(Chemo)enzymatic cascades – Nature’s synthetic strategy transferred to the laboratory
نویسندگان
چکیده
The astonishing efficiency with which living organisms build complex molecules from abundant starting materials has inspired chemists for centuries. Among the synthetic strategies that nature uses to achieve this efficiency, the combination of several enzymatic transformations in cascading sequences is of outstanding importance. With the rise of biocatalysis, researchers now have the tools at hand to mimic this strategy and develop artificial enzyme cascades of impressive complexity. This editorial review aims to introduce the reader to some key aspects of (Chemo)enzymatic Cascades, as well as to put the submissions to the present Special Issue into a broader perspective.

Assembly of cellulases with synthetic protein scaffolds in vitro
Background: Enzymatic cascades in metabolic pathways are spatially organized in such a way as to facilitate the flow of substrates. The construction of artificial cellulase complexes that mimic natural multienzyme assemblies can potentially enhance the capacity for cellulose hydrolysis. In this study, an artificial cellulase complex was constructed by tethering three cellulases to a synthetic p...
متن کاملWhy leave a job half done? Recent progress in enzymatic deracemizations
In recent years the usefulness of enzymatic systems to obtain a single stereoisomerically pure compound starting from a racemate has been expanded. Moreover, current advances in protein engineering, molecular biology and modeling tools are the basis to improve the catalytic properties of enzymes to face novel synthetic challenges. Also, medium engineering and novel immobilization methods of (bi...
متن کاملReinforcement learning based feedback control of tumor growth by limiting maximum chemo-drug dose using fuzzy logic
In this paper, a model-free reinforcement learning-based controller is designed to extract a treatment protocol because the design of a model-based controller is complex due to the highly nonlinear dynamics of cancer. The Q-learning algorithm is used to develop an optimal controller for cancer chemotherapy drug dosing. In the Q-learning algorithm, each entry of the Q-table is updated using data...
متن کاملp38 MAPK and PI3K/AKT signalling cascades in Parkinson’s disease
Parkinson's disease (PD) is a chronic neurodegenerative condition which has the second largest incidence rate among all other neurodegenerative disorders barring Alzheimer's disease (AD). Currently there is no cure and researchers continue to probe the therapeutic prospect in cell cultures and animal models of PD. Out of several factors contributing to PD prognosis, the role of p38 MAPKs (mitog...
متن کاملMimicking Biological Phenol Reaction Cascades to Confer Mechanical Function
Biotechnology enables the creation of proteinand nucleicacid-based materials to perform functions such as recognition, catalysis, signaling, and information storage/transfer. Interestingly, neither proteins nor nucleic acids are Nature’s most abundant biopolymers. Polysaccharides and polymeric phenolics (e.g., lignins) are more abundant. We contend that the reaction cascades that generate pheno...
